This is an oral tablet medicine; it is not a treatment for narcolepsy, which is a contraindication. Treatment planning should include the cause of continuing sleep problems.

Practical questions before taking it

MedlinePlus advises checking that a full night in bed is possible and reading the Medication Guide supplied with the prescription. Taking it with or soon after food can delay its effect. Persistent or worsening insomnia, or new changes in thoughts and behaviour, should be discussed with the prescriber rather than managed by increasing the amount taken.

Tell the healthcare team about alcohol use, other sleeping medicines, non-prescription products and supplements. Include relevant breathing problems, depression, pregnancy or breastfeeding. These details affect the assessment even if the medicine is taken only occasionally.

Safety and urgent warning signs

Somnolence is the main common adverse effect in the US label. Alertness, coordination and driving can remain impaired the next day. Alcohol and other central depressants add risk; the label advises avoiding alcohol and does not recommend combining Dayvigo with other insomnia medicines.

Sleepwalking, sleep-driving or other activities performed while not fully awake require stopping Dayvigo and contacting the prescriber immediately. Sleep paralysis, hallucinations and new mood deterioration also need assessment. Pregnancy data are inadequate to establish safety. Human milk transfer has been measured, but effects on breastfed infants are unknown; exposed infants need monitoring for excessive sedation.

Why the interaction check matters

In dedicated phase-one studies, itraconazole and fluconazole increased lemborexant exposure, whereas rifampin greatly reduced it. Lemborexant also reduced exposure to the measured bupropion component. These were pharmacokinetic studies, so their findings do not establish the safety of every medicine combination.

The US prescribing information advises avoiding strong or moderate CYP3A inhibitors and inducers. A pharmacist should check the complete regimen, including antifungals, antibiotics, seizure medicines and herbal products, before a new medicine is added.

Sources and scope

Dayvigo oral tablets: US prescribing information — Eisai; revised February 2025

Landry et al.: Lemborexant CYP3A and CYP2B6 interaction studies — Clinical Pharmacology in Drug Development 2021;10:681-690; DOI 10.1002/cpdd.915

MedlinePlus: Lemborexant patient information — US National Library of Medicine; revised 15 June 2023

Sources checked: 30 October 2026.
