Uses<\/h2>\n

Ambrisentan relaxes pulmonary blood vessels by blocking endothelin receptors. It treats pulmonary arterial hypertension (PAH), including use with other PAH treatment. The Turkish tablet information covers adults in functional classes II\u2013III. European information also includes selected children aged eight years and older; paediatric treatment requires a separate specialist assessment.<\/p>\n

Administration<\/h2>\n

This is an oral film-coated tablet, taken regularly as prescribed and swallowed whole. A PAH specialist decides whether it is used alone or with tadalafil. Do not add an erectile-dysfunction medicine or another pulmonary vasodilator on your own, even when some combinations are established treatment.<\/p>\n

Safety<\/h2>\n

Fluid retention and anaemia require monitoring. Liver tests are checked before treatment and followed during care. Severe liver impairment, marked baseline liver-enzyme elevation and idiopathic pulmonary fibrosis are important restrictions. Product-specific soya or other ingredient allergies also matter. Seek prompt assessment for rapid weight gain, worsening swelling, jaundice or increasing breathlessness.<\/p>\n

Interactions<\/h2>\n

Ciclosporin approximately doubles ambrisentan exposure, so the prescriber must limit the ambrisentan regimen and monitor the patient. Rifampicin can transiently increase exposure when started and warrants closer observation. Tadalafil combination treatment is supported by clinical evidence; it is not a contraindicated pairing, although oedema, anaemia and other adverse effects can be more frequent.<\/p>\n

Pregnancy and breastfeeding<\/h2>\n

Pregnancy and breastfeeding are contraindicated. A negative pregnancy test and reliable contraception are needed before treatment, with monthly testing during treatment. Suspected pregnancy requires immediate contact with the specialist to arrange an appropriate alternative.<\/p>\n

Adverse effects<\/h2>\n

Common effects include ankle swelling, headache, anaemia, nasal congestion, nausea and dizziness. Symptoms may overlap with PAH, so persistent or worsening problems need review rather than an unsupervised change in treatment.<\/p>\n
