It can reduce urgency, frequent urination and leakage associated with a sudden urge. Before treatment, a clinician should consider infection, obstruction, heart failure or kidney disease as alternative causes of urinary frequency.

Administration

The prolonged-release tablets are taken by mouth and swallowed whole with liquid, with or without food. Do not chew, divide or crush them. Treatment response and tolerability should be reviewed. Liver function and interacting medicines influence prescribing decisions.

Safety

Darifenacin must not be used with urinary or gastric retention, uncontrolled narrow-angle glaucoma, myasthenia gravis, severe hepatic impairment, severe ulcerative colitis or toxic megacolon. Caution is needed with significant bladder-outflow obstruction or severe constipation. Stop treatment and seek urgent care for swelling of the tongue or throat or trouble breathing. Inability to pass urine or severe abdominal symptoms needs prompt assessment.

Interactions

The Turkish and UK product information contraindicates potent CYP3A4 inhibitors, including ritonavir and itraconazole. This differs from US dose-limitation wording. Fluconazole and strong CYP2D6 inhibitors such as paroxetine or terbinafine call for cautious prescribing and reassessment. Ciclosporin and verapamil should be avoided under the UK product information. Rifampicin and carbamazepine may reduce effectiveness. Darifenacin can affect CYP2D6 substrates such as flecainide and increase digoxin exposure; monitoring is needed when treatment changes. Other bladder antimuscarinics and tricyclic antidepressants can intensify constipation, dry mouth or urinary retention.

Pregnancy and breastfeeding

Human pregnancy data are limited and use is not recommended during pregnancy. Passage into human milk is unknown. The decision about breastfeeding or treatment should consider both maternal benefit and potential infant risk.

Adverse effects

Dry mouth and constipation are particularly common. Headache, indigestion, abdominal discomfort and nausea also occur. Dizziness or visual changes can affect driving. Severe or persistent constipation should be discussed with the prescriber.

Sources

Regulatory prescribing information (English)

Turkish medical or product information (Turkish)

Additional verified references

FDA examples of drugs that interact with CYP enzymes and transporter systems

Xtandi (enzalutamide): EMA product information

Erleada (apalutamide): EMA product information

["post-12341","page","type-page","status-publish","hentry
["GET
