Current EU information includes high-risk early disease and advanced or metastatic disease. The Turkish Valamor document reviewed describes metastatic treatment with additional eligibility conditions; its indication should not be silently broadened to match the EU label.

Administration

Film-coated tablets are swallowed whole, with or without food, according to the oncology calendar. Treatment includes planned cycles and breaks. Do not crush, divide or take visibly damaged tablets. Endocrine treatment and ovarian suppression, where needed, have their own instructions. The team determines the regimen rather than using a generic schedule for every disease setting.

Safety

Ribociclib can prolong the heart’s QT interval. ECGs, electrolytes, blood counts and liver tests are needed. Palpitations, fainting or marked dizziness require urgent assessment. Report fever, jaundice, severe rash or new breathing difficulty promptly. Existing long QT, significant heart disease and abnormal electrolytes can increase risk and need specialist evaluation.

Interactions

Avoid other QT-prolonging medicines and strong CYP3A inhibitors whenever possible. Strong enzyme inducers can reduce efficacy. Ribociclib itself is a moderate-to-strong CYP3A inhibitor, with potency depending on the regimen: interactions should not be assigned the same strength at every dose. Certain statins, antipsychotics, erectile-dysfunction medicines and narrow therapeutic index substrates need avoidance or adjustment. Tamoxifen is not recommended as a partner because of additional QT concerns. Aromatase inhibitors are established partners.

Pregnancy and breastfeeding

Ribociclib is not recommended during pregnancy. Check pregnancy status before treatment and use effective contraception during treatment and for at least twenty-one days afterwards. Do not breastfeed during treatment or for at least twenty-one days after the final dose.

Adverse effects

Neutropenia, infections, nausea, fatigue, headache, leukopenia and abnormal liver tests are common. Diarrhoea and vomiting also occur, particularly in advanced-disease regimens. Report symptoms that interfere with drinking or eating.

Sources

Kisqali — EMA product information, Annex I SmPC

Turkish prescribing information: VALAMOR

Additional verified references

FDA examples of drugs that interact with CYP enzymes and transporter systems

Xtandi — EMA product information, Annex I SmPC

Erleada — EMA product information, Annex I SmPC

Ribociclib inhibits CDK4 and CDK6 and is combined with endocrine therapy for selected hormone receptor-positive, HER2-negative breast cancers.

["post-12403","page","type-page","status-publish","hentry
["GET
