Inclisiran: Uses, Interactions and Safety
Uses
Inclisiran is a small interfering RNA treatment that reduces production of PCSK9 in the liver, helping lower LDL cholesterol. It is used alongside diet and appropriate lipid treatment in primary hypercholesterolaemia or mixed dyslipidaemia. The Turkish LYPTIMZIA information covers adults; current European and US authorisations also include selected adolescents, which does not establish a Turkish paediatric indication.
Administration
A healthcare professional gives the subcutaneous injection. Initial treatment is followed by a further injection at three months and then six-monthly maintenance appointments. Keep those appointments even if you feel well. The clinic should decide how to handle a missed visit or a switch from another PCSK9 treatment.
Safety
Tell the team about previous allergy to inclisiran or its ingredients. Current US information includes serious hypersensitivity reports, including anaphylaxis and angioedema. Breathing difficulty or swelling of the face, tongue or throat requires urgent care. Dialysis timing also matters: European/Turkish information advises avoiding haemodialysis for at least 72 hours after administration.
Interactions
Statins are established background treatment, and no clinically meaningful interaction with atorvastatin or rosuvastatin is expected from the available data. Inclisiran is not expected to inhibit or induce major CYP pathways. This does not prove that every possible combination has been studied. Moving from evolocumab or alirocumab is a planned treatment transition; the label’s transition instructions do not support routine simultaneous use of both treatments.
Pregnancy and breastfeeding
As a precaution, use during pregnancy is preferably avoided because human data are limited. Human milk excretion is unknown, while animal milk excretion has been shown. Treatment and breastfeeding choices require an individual assessment.
Adverse effects
Injection-site pain, redness or rash are the main common reactions. Contact the clinic for severe, persistent or spreading local symptoms, and seek urgent help for systemic allergy symptoms.
